At long-term follow-up, CTO-NR patients had significantly higher rate of overall (p = .021) and cardiac mortality (p = .035) compared to those successfully revascularized. CONCLUSION In patients with systolic LV dysfunction (EF ≤ 45%), CTO revascularization was associated with significant lower rate of total and cardiac mortality compared to those with nonrevascularized CTO. © 2020 Wiley Periodicals, Inc.Opioid use by pregnant women is an understudied consequence associated with the opioid epidemic, resulting in a rise in the incidence of neonatal opioid withdrawal syndrome (NOWS) and lifelong neurobehavioral deficits that result from perinatal opioid exposure. There are few preclinical models that accurately recapitulate human perinatal drug exposure and few focus on fentanyl, a potent synthetic opioid that is a leading driver of the opioid epidemic. To investigate the consequences of perinatal opioid exposure, we administered fentanyl to mouse dams in their drinking water throughout gestation and until litters were weaned at postnatal day (PD) 21. Fentanyl-exposed dams delivered smaller litters and had higher litter mortality rates compared with controls. Metrics of maternal care behavior were not affected by the treatment, nor were there differences in dams’ weight or liquid consumption throughout gestation and 21 days postpartum. Twenty-four hours after weaning and drug cessation, perinatal fentanyl-exposed mice exhibited signs of spontaneous somatic withdrawal behavior and sex-specific weight fluctuations that normalized in adulthood. At adolescence (PD 35), they displayed elevated anxiety-like behaviors and decreased grooming, assayed in the elevated plus maze and sucrose splash tests. Finally, by adulthood (PD 55), they displayed impaired performance in a two-tone auditory discrimination task. Collectively, our findings suggest that perinatal fentanyl-exposed mice exhibit somatic withdrawal behavior and change into early adulthood reminiscent of humans born with NOWS. © 2020 Society for the Study of Addiction.The performances of routine tests such as FIB-4 and APRI in detecting cirrhosis and significant fibrosis in chronic hepatitis B (CHB) have been shown to be discrepant between studies.Novel testssuch asred cell distribution width-platelet ratio (RPR), γ-glutamyl transpeptidase to platelet ratio (GPR) andeasy liver fibrosis test (eLIFT) have also been introduced recently. To evaluate the aminotransferase influence on the performance of theseroutine tests, a total of 1005 CHB patients who underwent liver biopsies and routine tests were retrospectively analyzed.The diagnostic cutoffs referring to likelihood ratio were determined for excluding or including cirrhosis diagnosis, and also for ruling in significant fibrosis diagnosis.Theperformances ofRPR, FIB-4, eLIFT and APRI in detecting cirrhosis seemedimproved athigher ALTlevels, whistGPR was conversely impaired. The likelihood ratios of APRI 2 indiagnosing cirrhosisatALT less then 2 upper limit of normal (ULN), APRI 1.5 in determiningsignificant fibrosisatALT ≤5ULN andFIB-4 3.2 in diagnosing ≥Metavir F3 in the total cohort was∝, 14.6 and 20.6, respectively. The optimal cutoffs for cirrhosis diagnosis were increased with higher ALTs by testswhich included aminotransferaselevel, but not for RPR. The proportions of patients classifiedas having cirrhosis or nocirrhosis stratified by ALT level cutoffs were superior.Stepwise applying RPR, GPR and eLIFT would determine 60% of patients as having cirrhosis or nocirrhosis with an accuracy of 93.0%. In conclusion, the performance of aminotransferase comprisingtestsin detectingcirrhosis in CHB were influenced by ALT levels.Thus ALT stratified cutoffs may be a preferred alternative. All rights reserved.Lysine acylations, a family of diverse protein modifications varying in acyl group length, charge and saturation, are linked to many important physiological processes. Only a small set of substrate-promiscuous lysine acetyltransferases and deacetylases (KDACs) install and remove this vast variety of modifications. Engineered KDACs that remove only one type of acylation would help to dissect the different contributions of distinct acylations to cell physiology. Therefore, we developed a bacterial selection system for the directed evolution of KDACs with which we identified variants up to 400 times more selective for butyryl- compared to crotonyl-lysine. Structural analyses revealed that the enzyme adopts different conformational states depending on the type of acylation of the bound peptide. We used the butyryl-selective KDAC variant to shift the cellular acylation spectrum towards increased lysine crotonylation. Hence, these new enzymes will help dissecting the different roles of lysine acylations in cell physiology. © 2020 WILEY-VCH Verlag GmbH & Co. KGaA, Weinheim.BACKGROUND Prior studies have noted the importance of T cell immunoglobulin and mucin domain containing 4 (TIMD4) in various diseases and its functions on cell malignant behaviors. However, the biological function of TIMD4 in diffuse large B-cell lymphoma (DLBCL) is unknown. METHODS Relative expression of TIMD4 was analyzed based on the GSE56315 array including 88 cases of human tissues. TIMD4 expression in cells was detected using a quantitative reverse transcriptase-polymerase chain reaction and western blot experiments. Cell proliferation was measured using the cell counting kit-8 (CCK-8) assay and apoptotic properties were assessed through the detection of related proteins by western blotting. The underlying molecular mechanism of TIMD4 in DLBCL was predicted and confirmed using KEGG enrichment analysis and western blotting. RESULTS The results indicate that TIMD4 is overexpressed in DLBCL tissues and the poor prognosis of DLBCL patients is significantly linked with the higher TIMD4 expression. The loss-of-TIMD4 experiment in CYP6D reveals that knockdown of TIMD4 blocks cell growth and accelerates cell apoptosis, whereas the gain-of-TIMD4 experiment in Raji cells suggests that up-regulation of TIMD4 promotes cell proliferation and inhibits cell apoptosis. The activity of the Wnt/β-catenin pathway is mediated by the TIMD4 expression in DLBCL cells. CONCLUSIONS These findings demonstrate that TIMD4 is up-regulated in patients with DLBCL and the regulatory effects of TIMD4 on cell proliferation and apoptosis are associated with the Wnt/β-catenin pathway, posing a novel target for DLBCL therapy. © 2020 John Wiley & Sons, Ltd.Coronary artery dissection is a well-known complication of percutaneous coronary interventions. In this setting, coronary artery dissection is caused by mechanical injury of the arterial wall. However, dissection may also occur spontaneously. In both situations, an intimal tear or dissection allows blood entering and separating the layers of the coronary arterial wall. Despite percutaneous approach is still the preferred and the quickest way to restore coronary flow, it remains challenging. One of the main reasons for procedural failure is due to the difficult advancement of a guidewire into the true lumen. In such situation, the ideal wire should have a soft tip, high torque control, and excellent flexibility. Assuming that, the „new rope coil” composite core Suoh 0.3 guidewire, with its unique combination of characteristics, could allow better orientation insight into a dissected coronary artery increasing the chance of procedural success. The reactivity of pentaphosphaferrocene, [Cp*Fe(η5-P5)] (Cp* = C5Me5), with two different low-valent aluminium compounds was investigated. The steric and electronic environment around the [AlI] centre was found to be very crucial for the resulting Al-Fe polyphosphides. Reaction with the sterically demanding [Dipp-BDIAlI] (Dipp-BDI = [2,6-iPr2C6H3NCMe]2CH-) resulted in the first Al-based neutral triple-decker type polyphosphide complex. In case of [(Cp*AlI)4], an unprecedented regioselective insertion of three [Cp*AlIII]2+ moieties into two adjacent P-P bonds of the cyclo-P5 ring of [Cp*Fe(η5-P5)] was observed. KGaA, Weinheim.Here we propose an innovative strategy to synthesize single-crystal nanowires (NWs) of the Al 3+ dicarboxylate MIL-69(Al) MOF by using graphene oxide nanoscrolls as structure directing agents. MIL-69(Al) NWs with an average diameter of 70 ± 20 nm and lengths up to 2 μm were found to preferentially grow along the [001] crystallographic direction. Advanced characterization tools (electron diffraction, TEM, STEM-HAADF, SEM, XPS) and molecular modelling revealed the mechanism of formation of MIL-69(Al) NWs involving size-confinement and templating effects. The formation of MIL-69(Al) seeds and the self-scroll of GO sheets followed by the anisotropic growth of MIL-69(Al) crystals are mediated by specific GO sheets/MOF interactions. As an innocent oxidizing agent it possesses a strong and fully reversible half-wave potential vs. Fc + /Fc in coordinating MeCN ( E °’ = 1.21 V), but also in almost non-coordinating o DFB (= 1,2-F 2 C 6 H 4 ; E °’ = 1.29 V). It allows for the oxidation of [Fe III Cp* 2 ] + to [Fe IV (CO)Cp* 2 ] 2+ and [Fe IV (CN- t Bu)Cp* 2 ] 2+ in common laboratory solvents and is compatible with good σ-donor ligands like L = trispyrazolylmethane to generate novel [M(L) x ] n + complex salts from the respective elemental metals. © 2020 WILEY-VCH Verlag GmbH & Co. KGaA, Weinheim.BACKGROUND Determination of protective and risk factors associated with Internet Gaming Disorder (IGD) are among the most important pathways to develop prevention strategies for IGD. Previous research has shown that familial factors are associated with IGD. In our study we aimed to assess the parental attitude of adolescents with IGD and investigate psychiatric comorbidity. METHODS We assessed family structure, family relationship, parental attitude (in a bi-directional assessment), and psychiatric comorbidity in 50 adolescents aged 12-18 years who meet DSM 5 criteria for IGD in comparison with control group. Parental attitudes are assessed with Parental Attitude Research Instrument (PARI) (filled by mother) and Parenting Style Inventory (PSI) (filled by adolescents). RESULTS Our findings suggest that according to mothers’ opinions there was no significant difference in the subscale scores between the IGD and the control group. On the other hand, acceptance-involvement and psychological autonomy subscale scores of the PSI filled by adolescents were found to be significantly lower in the IGD group.
