    The concentrations of insulin, insulin-like growth factor 1, leptin, adiponectin, haptoglobin, glucose, and revised quantitative insulin sensitivity check index did not differ between the groups, whereas serum concentrations of glycerophospholipids were lower before calving in HBCS-PH than in HBCS-PN cows. Glycine was the only amino acid that had higher concentration after calving in HBCS-PH than in HBCS-PN cows. The circulating concentrations of some short- (C2, C3, and C4) and long-chain (C12, C160, C180, and C181) acylcarnitines on d 21 pp were greater in HBCS-PH than in HBCS-PN cows, indicating incomplete FA oxidation. In conclusion, the use of ML approaches involving data from targeted metabolomics in serum is a promising method for differentiating divergent metabotypes from apparently similar BCS phenotypes. Further investigations, using larger numbers of cows and farms, are warranted for confirmation of this finding.The objective of this study was to investigate whether cultured ruminal epithelial cells (REC) responded to lipopolysaccharide (LPS) stimulation and determine whether LPS induced a proinflammatory response. Primary bovine REC were isolated and grown in culture for 2 studies. In study 1, REC were isolated from Holstein bull calves (n = 8) and grown in culture for 10 to 12 d. Cells were then exposed to 0, 10,000, 50,000, or 200,000 endotoxin (E)U/mL of LPS (Escherichia coli O55B5) for either 6 or 24 h. The effect of LPS exposure on cell viability was analyzed by flow cytometry using a propidium iodide stain. In study 2, cells were isolated from Holstein bull calves (n = 5) and yearling beef heifers (n = 4). Cells were exposed to either 1,000 or 50,000 EU/mL of LPS using the following conditions (1) medium alone time-matched controls, (2) 12-h LPS exposure, (3) 24 h of LPS exposure, (4) 36 h of LPS exposure, (5) 12 h of LPS exposure followed by LPS removal for 24 h before restimulating with LPS for an additionalssion was significantly greater after a single 12 h of LPS exposure than after RPT exposure, suggesting repeated exposure of REC to LPS may induce a tolerogenic effect. When LPS was removed from the medium (RVY), transcript abundance for all genes analyzed decreased and expression of TLR2, TLR4, and IL7 returned to baseline levels, suggesting REC recovered following exposure to LPS. Overall, the data suggest cultured REC respond to LPS stimulation by increasing transcription of proinflammatory genes and this transcriptional response was influenced by the dose, duration, and frequency of LPS exposure.The objective of this study was to describe the prevalence and trends in antimicrobial resistance for bacterial pathogens associated with bovine respiratory disease (BRD) isolated from samples submitted to the Wisconsin Veterinary Diagnostic Laboratory (WVDL). Data were retrospectively collected from bovine respiratory isolates including Pasteurella multocida, Mannheimia haemolytica, Histophilus somni, and Bibersteinia trehalosi identified at the WVDL between January 2008 and December 2017. Antimicrobial susceptibility testing data were queried from antimicrobial resistance databases at the WVDL. A total of 4,261 isolates were identified. Pasteurella multocida was most frequently identified, accounting for 2,094 isolates (49% of total) over the study period. Mannheimia haemolytica was the second most frequently isolated bacterial respiratory pathogen (n = 1,267, 30%) followed by H. somni (n = 749, 18%) and B. trehalosi (n = 151, 4%). Over the 10-yr period, B. trehalosi had the highest median percentage of isoware of the bacterial pathogens most commonly associated with BRD and work toward early disease detection, proper antibiotic administration, and monitoring lung lesions to ensure that their treatment protocols improve lung health.Lameness is a major welfare problem in the dairy industry. Environmental factors, such as flooring surface, as well as cow-level factors, such as udder fill, can influence gait. The aim of the current study was to test whether motivation to walk affects gait attributes and whether this effect differs between lame and sound cows. We trained cows to walk down an alley for a food reward and assessed walking speed, stride length, head bob, and back arch of cows previously identified as either lame (n = 7) or sound (n = 10). Cows were assessed when they walked toward a food reward and toward no reward. Cows walked faster and had longer stride length and less variation in head bob when approaching the reward; these effects were similar in both sound and lame cows. We concluded that motivation to walk affects several gait attributes of dairy cows.Drying-off, calving, and start of lactation are critical transition events for a dairy cow. As a consequence, most animal health issues occur during these periods. By extending the voluntary waiting period for first insemination after calving, calving interval (CInt) can be extended, with possible positive effects for fertility and health. Some cows might be better suited for an extended CInt than others, due to differences in milk yield level, lactation persistency, or health status, which would justify a customized CInt based on individual cow characteristics. This study aims to investigate 13 farms with customized CInt, with respect to calving to first service interval (CFSI), accomplished CInt, services per conception (SC), conception rate at first artificial insemination (CR1AI), peak yield, lactation persistency, 305-d yield, and effective lactation yield. In total, 4,858 complete lactations of Holstein Friesian cows between 2014 and 2019 from the 13 farms were grouped by parity (1 or 2+) and CFSI (CFSI class; CFSI-1 364 d), showing that these cows were not able to conceive for the desired CInt. More than 50% of cows in CInt classes 3 and higher (CInt ≥ 420 d) had an earlier first insemination before successful insemination (CFSI class 1; less then 196 d), showing that these extended CInt classes consisted of both cows with an extended waiting period for first insemination and cows that failed to conceive at earlier insemination(s). On most farms, lactation persistency was greatest in CInt class 1 ( less then 364 d), probably related to the low peak yield in this class. When this shortest CInt class was excluded, persistency increased with extended CInt classes on most farms. Although at the majority of farms 305-d yield was greater in CInt ≥ 532 d, effective lactation yield at most farms was greatest in CInt from 364 to 531 d, especially for multiparous cows. Based on the results of this study, a CInt between 364 and 531 days seems most optimal for milk production, when high-yielding cows were selected.Antibody-detecting tests for Mycobacterium avium ssp. paratuberculosis (MAP) have low sensitivity and imperfect specificity for detection of infection. Sensitivity increases as the disease progresses. Aside from infection status and stage of disease, several factors affect test performance. These factors have not yet been studied in dairy cows producing lower volumes of milk with higher solids concentration, such as those managed in low-input, pasture-based production systems. Furthermore, the effect of correcting for these associations on individual and herd test status is also unknown. The first objective of this study was to examine the relationship between MAP antibody response in milk and milk yield, somatic cell count (SCC), fat and protein contents, and stage of lactation in dairy cows enrolled in the national Johne’s Disease Control Programme (JDCP) in Ireland. The second objective was to examine the effect of correcting the antibody response for these associations on the test status of individual cowiations, 93 multiparous cows and 20 primiparous cows changed category (negative, suspect, or positive). When considered at the herd-test level, out of a total of 531 herd tests, 1 herd changed from negative to positive, and 5 herds changed from positive to negative. This study provides useful information to aid in the interpretation of antibody results for herds testing animals for the presence of MAP infection. At an overall population level, correction of the serological response for non-disease-associated factors has the potential to change the status of only a small number of cows. At the herd level, the proportion of herds changing status was minimal. However, depending on the implications of a herd-level serological diagnosis, consideration should be given to correcting for these non-disease-associated variables within the context of national JD control programs.In dairy cattle, selecting for lower methane-emitting animals is one of the new challenges of this decade. However, genetic selection requires a large number of animals with records to get accurate estimated breeding values (EBV). Given that CH4 records are scarce, the use of information on routinely recorded and highly correlated traits with CH4 has been suggested to increase the accuracy of genomic EBV (GEBV) through multitrait (genomic) prediction. Therefore, the objective of this study was to evaluate accuracies of prediction of GEBV for CH4 by including or omitting CH4, energy-corrected milk (ECM), and body weight (BW) as well as genotypic information in multitrait analyses across 2 methods BLUP and single-step genomic BLUP (SSGBLUP). A total of 2,725 cows with CH4 concentration in breath (14,125 records), BW (61,667 records), and ECM (61,610 records) were included in the analyses. Approximately 2,000 of these cows were genotyped or imputed to 50K. Ten cross-validation groups were formed by randomly grouing information on both traits (BW and ECM) increased the accuracy by up to 90%. Scenarios that did not include CH4 in the reference population had the lowest correlations (0.17-0.33) with single-trait CH4 GEBV, and scenarios with CH4 in the reference population had the highest correlations (0.41-0.81). Thus, failure to include CH4 in future reference populations results in predicted CH4 GEBV, which cannot be used in practical selection. Therefore, recording CH4 in more animals remains a priority. Finally, multiple-trait genomic prediction using routinely recorded BW and ECM leads to higher prediction accuracies than traditional single-trait genomic prediction for CH4 and is a viable solution for increasing the accuracies of GEBV for scarcely recorded CH4 in practice.Saturated fatty acid supplements commonly fed to dairy cows differ in their fatty acid (FA) profile. Some supplements with very high enrichments of palmitic acid (PA) or stearic acid (SA) have been reported to have low total-tract digestibility. Saturated FA have the potential to form crystalline structures at high purity that may affect digestibility. Differential scanning calorimetry (DSC) is a thermal technique commonly used in materials science to measure the change in heat flow as energy is absorbed or released from a sample during heating, and it was used to characterize a series of experimental and commercial fat supplements. Our hypothesis was that products with very high enrichment of either PA or SA would differ in thermal characteristics compared with those that include moderate levels of a second FA because of the formation of secondary crystalline structures, which may contribute to decreased digestibility. First, replicated runs demonstrated low variation in melting temperature (MT) and enthalpy (coefficient of variation 98%) had the highest MT, and one moderately enriched in PA (∼85%) that contained 10.
